Summary
TFC1 encodes one of six subunits of RNA polymerase III (pol III) transcription factor TFIIIC, which is composed of Tfc1p, Tfc3p, Tfc4p, Tfc6p, Tfc7p, and Tfc8p. Genes that are transcribed by pol III, all encoding small RNAs such as tRNAs, 5S rRNA genes, and U6 spliceosomal RNA, contain two conserved promoter elements termed the A and B boxes. The A box is bound by the TA subcomplex of TFIIIC, and the B box is bound by the TB subcomplex. Tfc1p is part of the TA subcomplex of TFIIIC, along with Tfc4p and Tfc7p. Tfc1p is important for recognition of and binding to the A box, and is required for pol III start site selection. During pol III transcription initiation, TFIIIC first binds to DNA and then recruits the subunits of TFIIIB: Brf1p, Bdp1p, and Spt15p (also known as TATA-binding protein). The major role of TFIIIC is to facilitate the assembly of TFIIIB, which is a requirement for pol III transcription initiation. TFIIIC is also involved in transcription reinitiation by pol III, and its binding to chromatin may have a regulatory role in the determination of chromatin structure.
